Defeat the Inquest champion takes place in front of the Asura Gate in Rata Arcanum.

In order to advance the dialogue and get the Champion to spawn, kill all of the inquest in the area (you might have to wait for some to re-spawn), when dropped, grab any Inquest keypads and disable the closest terminal showing a wrench.
After you've disabled 8 of 8 control panels, the Champion will appear.
